Product Description:

SS-31 (also elamipretide) is a synthetic tetrapeptide (D-Arg-2′,6′-dimethylTyr-Lys-Phe-NH₂). It concentrates in the inner mitochondrial membrane. The FDA-approved drug form is Forzinity (elamipretide hydrochloride), labeled only for Barth syndrome.

How It Works

SS-31 binds cardiolipin, the phospholipid that holds mitochondrial cristae and the electron-transport chain in place. That interaction helps stabilize membrane structure under stress, keep cytochrome c working as an electron carrier rather than a peroxidase, improve ATP production and coupling, and cut electron leak and ROS at the source. It can also affect membrane surface charge and proteins involved in ADP/ATP handling (including ANT). Effects show up mainly in damaged or aged mitochondria; healthy mitochondria usually change little. It is structural support for energy production, not a stimulant.

Potential Benefits and Why People Use It

In fitness, longevity, and regenerative circles it is used for mitochondrial energy, recovery, and age-related decline. Animal work reports better muscle ATP output, exercise tolerance, and protection in ischemia, heart, kidney, and some neurologic injury models. The approved human use is narrower: Forzinity received FDA accelerated approval in 2025 to improve muscle strength in Barth syndrome patients weighing at least 30 kg. Other programs (primary mitochondrial myopathy, heart failure, dry AMD) have not produced approved indications; several late-stage trials missed primary endpoints.

Important Caveats

  • Evidence: Strong preclinical mechanism; limited and mixed human data outside Barth syndrome. Broad anti-aging, performance, or recovery claims are unproven.
  • Regulation: Approved only as Forzinity for that rare-disease indication. Research/gray-market SS-31 is not the approved product and quality varies. Not a general supplement. Not named on the WADA list, but unapproved peptide use can fall under S0.
  • Safety: Most common issues in trials are injection-site reactions; headache, dizziness, nausea, or fatigue also occur. Long-term safety in healthy users of non-pharmaceutical peptide is not established. Seek qualified medical advice before use.
